Celentyx is proud to have contributed human translational evidence supporting P2X7 receptor antagonism as a powerful therapeutic strategy to combat neuroinflammation (https://doi.org/10.1093/brain/awag068).
Published in Brain, this research provides key evidence that targeting the human purinergic P2X7 receptor can limit hostile, pathology-driven pro-inflammatory responses in human models of traumatic brain injury (TBI). Through a rigorous pharmacological investigation leveraging Celentyx’s advanced Neuroinflammation Platforms, including a human cellular microglial model alongside human functional brain slices, the study demonstrates the translational potential of the clinical-stage, CNS-penetrant P2X7 receptor antagonist, JNJ-54175446, to reduce secretion of relevant neuroinflammatory cytokines. These findings mark an important step forward in understanding how P2X7 receptor antagonism may be harnessed to modulate microglia-driven inflammation following TBI. More broadly, they strengthen the scientific rationale for investigating this pathway across neurological and psychiatric diseases in which dysregulated neuroinflammation is implicated. At Celentyx, we are committed to accelerating the discovery and development of therapies for diseases mediated via the immune system. Our extensive portfolio of human advanced in vitro models enables investigation of therapeutic mechanisms and evaluate drug candidates targeting immune system pathways, including those relevant to human microglia and neuroinflammation.
