Phosphoflow Assays

Phosphoflow enables quantification of phosphorylated signalling molecules in immune cell subsets. This powerful technique enables a quantitative assessment of test drug impact as well as defining which cells are affected. We frequently use phosphoflow to assess target proximal impact of test drug in screening or biomarker assays including target engagement notification in samples from clinical trials.

The ‘Example Response’ pseudocolour plots show typical responses of a responding and non-responding cell type from 
a investigation of ten immune cell types in a PBMC mix identified by expression of phenotypic markers. 

​Subsequent testing across multiple donors demonstrates reproducibility of response (bottom left), as well as the pharmacological impact and characterisation of two JAK inhibitors in clinical use (bottom right).
