Assays for assessing cytokine production from immune cell subsetsCelentyx offer intracellular cytokine staining to enable flow cytometric quantification of cytokine production at the single-cell level. This allows subsets of immune cells to be identified that may be responsible for effector function in immuno-oncology or responsible for tissue damage in autoimmunity and inflammatory disease. In the example PBMC sample below, cells were stimulated with a CD8-specific peptide pool, and IFNγ and TNFα production from the PD-1¯CD137¯, PD-1+CD137¯ and PD-1¯CD137+ populations analysed. The PD-1+CD137¯ population produces little IFNγ and TNFα, whereas the majority of the PD-1¯CD137+ produces IFNγ and TNFα.
Assays may be suitable for evaluating modulators of CD3, CD27, CD28, CD40, CD40L, CD70, CD80, CD86, CTLA-4, JAK1, JAK2, JAK3, ICOS, ICOSL, IL-2R, IL-9R, IL-21R, OX40, OX40L, PD-1, PDL-1, TIGIT, TIM-3, STAT1, STAT3, STAT5 and others. |
